Kudirat Kekere-Ekun began her early education at St. Mary’s School in Lagos and continued her secondary education at Queen’s College, Yaba, Lagos. In 1980, she obtained her Bachelor of Laws (LL.B.) degree from the University of Lagos. She completed her mandatory one-year program at the Nigerian Law School and was called to the Nigerian Bar in 1981. To further her education, she pursued a Master of Laws (LL.M.) degree at the London School of Economics and Political Science, graduating in 1983.

Kudirat Kekere-Ekun started her legal career as a State Counsel at the Lagos State Ministry of Justice in 1982. She later joined the Lagos State Judiciary as a Senior Magistrate Grade II in December 1989. In July 1996, she was appointed as a Judge of the High Court of Lagos State. She also served as the Chairman of the Robbery and Firearms Tribunal, Zone II, Ikeja, Lagos, from November 1996 to May 1999.

In September 2004, she was elevated to the Court of Appeal, where she served in various divisions and as the presiding justice in the Makurdi and Akure divisions. On July 8, 2013, she was appointed as a Justice of the Supreme Court of Nigeria, becoming the fifth female to hold this position.

Her notable rulings include being part of the seven-member panel that decided the Imo State governorship case in 2020, which resulted in the sacking of Emeka Ihedioha and the declaration of Hope Uzodinma as the winner. On August 23, 2024, she was sworn in as the Chief Justice of Nigeria by President Bola Ahmed Tinubu, making her the first woman from the Southwest geopolitical zone to achieve this role.
